RESULTS: Horizontal, vertical, stereotypic activity levels and total distance were shown significant differences
according to both groups and gender. Particularly stereotypic activities were found significantly lower (p<0.001)
in females reared in EC or IC in comparison to males. In forced swim test also, depression parameters of
females reared in EC were found significantly reduced (p<0.01); but males were found more susceptible to
depression and more quickly learned helplessness. While anxiety levels of females reared in different conditions
were not significantly different; in males reared in EC, duration of stay in the closed arm were found
significantly longer (p<0.05) and total number of entries were found significantly lower (p<0.001) than those
reared in SC.
CONCLUSION: Behavioral parameters of pups reared in diverse conditions following exposure to stress during
prenatal period were shown significant gender-dependent differences. While enrichment of environmental
conditions leads to negative effects in males, in terms of vulnerability to depression and anxiety; in females, in
contrary, causes positive effects.

AMAÇ: Otuz dakikadan uzun süren konvülziyon ya da aynı süre içinde bilinç yerine gelmeden devam eden seri
konvülziyonlar status epileptikus (SE) olarak adlandırılır. SE gerek hayatı tehdit etmesi, gerekse de ciddi
sorunlar oluşturması nedeniyle hızla tanınıp tedavi edilmesi gereken bir tablodur. Yeni nesil antiepileptik ilaçlar
arasında yer alan Levetirasetam’ın klinikte SE olgularında kullanımı ve etkilerine ait oldukça az veri
bulunmaktadır. Sunulan çalışmada immatür hayvanlarda kainik asit (KA) ile status epileptikus modeli
oluşturulurak Levetirasetam’ın öğrenme, hafıza ve davranış üzerine etkileri değerlendirilmiştir.
GEREÇ ve YÖNTEM: Araştırmada 21 günlük 23-50 gr arasında toplam 24 adet erkek, Spraque-Dawley sıçan
kullanıldı. Bunlardan 15 tanesine tek doz 10 mg/kg Kainik asit (Sigma) intraperitoneal olarak verildi ve SE
oluşumu gözlendi. Diğer 9 tanesine tek doz KA sonrası 14 gün boyunca 200 mg/kg dozunda Levetirasetam
(Keppra) intraperitoneal olarak verildi. SE’den iki hafta sonra uzamsal öğrenme Morris su tankı, emosyonel
öğrenme T labirent testi ile davranış değişiklikleri ise açık alan düzeneğinde değerlendirildi.
BULGULAR: Veriler Independent T Testi’nde analiz edildi. Açık alan testinde, yükseltilmiş T labirent testinde ve
Morris su tankında gruplar arasında anlamlı bir fark bulunamadı (p>0.05).
SONUÇ: KA ile oluşturulan SE modelinde levetirasetamın davranış, öğrenme ve hafıza parametreleri üzerinde
etkili olmadığı saptanmıştır.

The effects of levetiracetam on learning, memory and behavior in rats with
kainic acid–induced status epilepticus
OBJECTIVES: Status epilepticus (SE), is currently defined as a single clinical seizure lasting more than 30 mins
or repeated seizures over a period of more than 30 mins without intervening recovery of consciousness. SE
need to be life threatening, as well as to create serious problems because of an entity that must be rapidly
recognized and treated. There is very little data about the new generation of antiepileptic drugs and the effects
of the use of the levetiracetam clinical cases of SE. In this research, kainic acid (KA)-induced SE was formed in
immature rats to evaluate the effects of levetiracetam on learning, memory and behavior.
MATERIALS & METHODS: Experiments were carried out on male Spraque-Dawley rats, 21 days old, weighing
23–50 g. KA-induced SE rats (15 rats) were injected with 10 mg/kg intraperitoneal kainic acid and rats were
observed. For 14 days, nine rats received 200 mg/kg Levetiracetam (Keppra) after KA-induced SE. Two weeks
after SE, spatial learning was evaluated with Morris water maze, emotional learning was evaluated with T maze
and behavioral changes was evaluated with open field.
RESULTS: Data were analyzed with Independent T Tests. No differences were found on open field area tests,
elevated T maze and Morris water maze between groups (p>0.05).
CONCLUSION: In conclusion, in kainic acid-induced SE model, levetiracetam had no effect on behavior,
learning and memory parameters.

AMAÇ: Çalışmamızda, stresin Morris su tankında spasyal bellek performansına etkisinin olup olmadığını
göstermeyi amaçladık.
GEREÇ ve YÖNTEM: Çalışmada 22 adet Wistar Albino cinsi sıçan kullandık. Sıçanlar kontrol (n=10) ve stres
(n=12) olmak üzere 2 gruba ayrıldı. Bellek performansı Morris su tankında uzak hafıza deneyi ile değerlendirildi.
Stres uygulaması su tankı deneylerinden önce her hayvana 10 dakika hareketsizlik stresi uygulanarak yapıldı.
Sıçanların uzak hafıza deneyindeki ilk dört gün atışlarında platformu bulma süreleri, beşinci gün atışında ise 1
dk boyunca kadranlarda geçirdikleri süreler kaydedildi. Süre verilerinin normal dağılıma uygunluğunu test
etmek için Shapiro-Wilk normal testi uygulandı. Kontrol ve stres gruplarını her gün için olan süre değerleri
açısından karşılaştırırken Student t ve Mann Whitney U testleri kullanıldı. Günler arasında süre değerleri
açısından yapılan karşılaştırmalarda ise Tekrarlanan Ölçümlü Anova testi ve bu teste ait ikili karşılaştırmalar için
de LSD testi uygulandı.
BULGULAR: Hem kontrol grubunda hem de stres grubunda 1. gün atışı ile 3. ve 4. gün atışları arasında
platformu bulma süreleri açısından anlamlı fark bulunurken (kontrol grubu için sırasıyla p=0.001, p<0.001,
stres grubu için sırasıyla p<0.001, p<0.001) iki grupta da 1. gün ve 2. gün atışlarında platformu bulma
sürelerinde fark gözlenmiyordu. Gruplar arasında günler açısından anlamlı bir fark yoktu. 5. gün atışında ise
hem kontrol grubunda hem de stress grubunda kadranlarda geçirilen süre açısından anlamlı fark bulundu
(sırasıyla p<0.001, p=0.27).
SONUÇ: Hareketsizlik stresi, Morris su tankında sıçanların hem ilk dört günlük atışlarda platformu bulma
sürelerini hem de 5. gün atışlarında doğu kadranında geçirdikleri süreleri etkilemedi.

Does stress affect spatial memory performance in Morris water maze?
OBJECTIVES: We aimed to investigate if stress has an effect on spatial memory in Morris water maze.
MATERIALS & METHODS: Twenty two Wistar Albino types of rats were used in our study. Rats were divided
into 2 groups as control (n=10) and stress (n=12). Memory performance was evaluated using long term
memory experiment in Morris water maze. Stress was applied to each rat before water maze test for 10
minutes by applying immobilization stress. For the first 4 days in the long term memory experiments of rats,
time to find platform and at fifth day, time spent at quadrant was recorded. Shapiro-Wilk normality test was
used to test the normal distribution of the data. Student t and Mann Whitney U test were used in comparing
data of control and stress groups for time spent each day. Repeated measures analysis of variance was applied
in comparison of time spent among days and LSD was used to test pair comparisons.
RESULTS: Both in control and stress group, significant difference was found between 1st and 3rd and 1st and
4th day in the time spent to find the platform (control group p=0.001, p<0.001, respectively, stress group
p<0.001, p<0.001, respectively). There was no significant difference between 1st and 2nd day in the time to
find the platform in both groups. On 5th day, a significant difference was found in both control and stress
groups in the time spent on quadrant (p<0.001, p=0.27, respectively).
CONCLUSION: Immobilization stress did not affect the time to find platform in the first 4 days and the time
spent on east quadrant in 5th day in Moris water maze.

Hayvanlarda öğrenme kapasitesi ile hipokampustaki CaMIIα ve pCaMIIα
seviyeleri arasındaki korelasyonun genç Long Evans sıçanlarda araştırılması

AMAÇ: Sunulan çalışmanın amacı davranış testindeki performanslarına göre “iyi” ve “kötü” öğrenen olarak
sınıflandırılan genç erkek Long Evans sıçan populasyonunda kısa ve uzun süreli bellekteki bireysel varyasyonun
CaMKIIα ve fosforile formu pCaMKII proteininin hipokampustaki seviyesine bağlı olup olmadığını incelemektir.
GEREÇ ve YÖNTEM: Sıçanlar kısmen pekiştirilmiş radyal labirentteki performanslarına göre “iyi” ve “kötü”
öğrenen olarak sınıflandırılmıştır. “İyi” ve “kötü” öğrenen sıçanların hipokampuslarındaki CaMKIIα ve pCaMKII
proteinlerinin seviyesindeki farklılıkları belirlemek için Western Blot tekniği uygulanmıştır.
BULGULAR: Çalışmadan elde edilen sonuçlara göre hipokampus beyin bölgesine bağlı olarak bilinen mekansal
öğrenme testinde çok iyi ve çok kötü performans gösteren sıçanlar arasında hipokampustaki CaMKIIα’nın
ekspresyonunda (toplam CaMKIIα konsantrasyonuna bakılınca) veya aktivasyonunda (fosforile edilen CaMKIIα
konsantrasyonuna bakılınca) fark görülmemiştir.
SONUÇ: Daha önce yapılan birçok araştırmada farmakolojik ve/veya genetik yöntemlerle CaMKII’nin
ekspresyonunun veya aktivitesinin bastırılmasının hem hücresel (LTP) hem de davranışsal belleğin oluşumunu
engellediği gösterilmiştir. Bu negatif sonucun birkaç sebebi olabilmektedir. İlki, eğer öğrenme eğitiminin
tetiklediği CaMKII’nin aktivasyonunun sadece hipokampusun belli bir hücre kitlesine sınırlandırılmış olduğunu
varsayarsak, tüm bölgeden alınan protein ekstrakt üzerinde yapılan Western blot analizinde bu değişiklik
kamufle edilmiş olabilir. Diğer taraftan, daha önce yapılan bazı çalışmalarda, CAMKII aktivasyonunun öğrenme
sürecinin erken safhalarında önemli olduğu gösterilmiştir. Ancak, mevcut deneylerde uygulanan eğitim
modelinde uzun süreli bellek izlerinin oluşmasının yanı sıra her oturumda kısa süreli belleğin çalıştırılması
gerekmekteydi ve “kötü” öğrenen sıçanlar “iyi” öğrenenlere nazaran hem uzun hem de kısa süreli bellek hata
sayısında anlamlı bir artış sergiliyordu. Alınan sonuçlar, CAMKII’nin kısa süreli bellek mekanizmasında önemli bir
rol oynamadığına işaret etmektedir.

Study of the relation between the animals’ learning capacity and the
hippocampal levels of CaMIIα and pCaMIIα in random population of Long Evans
rats
OBJECTIVES: The aim of the present study is whether individual variations in long-term and short-term
memories of rats classified as “good” and “poor” learners according to their performance in behavioral task is
related with expression of CaMKIIα and its phosphorylated form (pCaMKII) in hippocampus.
MATERIALS & METHODS: Young male Long-Evans rats were classifed as “good” and “poor” learners based on
their performances in partially baited radial arm maze. The molecular levels of CaMKIIα ve pCaMKII proteins in
the hippocampus were determined by Western Blotting.
RESULTS: There is no significant difference in hippocampal CaMKIIα expression or activation (compared to
pCaMKIIα concentration) between the rats showing good and poor performance in spatial learning task.
CONCLUSION: In previous studies, it is showed that the decrease of expression or activation of CaMKIIα
inhibit the formation of both cellular (LTP) and behavioral memory. However, in our study, if it is assumed that
CaMKII activation triggered by learning task is restricted to a region of given cells in the hippocampus, the
effect of activation may be camouflaged in Western blot analysis done on the whole hippocampus extract. Also,
as previously shown that CaMKII activation is important in the early phases of learning. However, in the
learning task applied in present study, besides the formation of long-term memory traces, the rats should use
short-term memory in each trial. Furthermore, “poor” learners were showing significant increase in the number
of errors both in long-term and short-term memories. In summary, our results indicate that CaMKII does not
have important role in short-term memory mechanisms.

AMAÇ: Susamış deneklerde içecek tüketimi davranışının emosyonel kognitif içerikli bir uyaranın (gülen yüz)
bilinçaltında işlenmesi ile değiştirilip değiştirilemeyeceğini araştırmak.
GEREÇ ve YÖNTEM: Deneye 27 sağlıklı denek, dokuzar kişilik 3 ayrı grup oluşturularak katılmıştır. Görsel
uyaranların gruplara dağılımı şöyledir: 1. Kontrol grubu: 16 ms boyunca nötr yüz, 400 ms boyunca yine nötr
yüz 2. Çok kısa süreli bilinçaltı grubu: 16 ms boyunca gülen yüz, 400 ms boyunca nötr yüz 3. Uzun süreli
bilinçaltı grubu: 16 ms boyunca nötr yüz, 400 ms boyunca gülen yüz. Resimler toplam 15 kere gösterilmiş,
deneklerden sadece resimdeki kişinin cinsiyetini belirtmeleri istenmiştir. Deneklere deney öncesinde 6 tane
baharatlı kraker yedirilmiş, yapılan anket hepsinin susamış olduğunu göstermiştir. Deney sonrasında 25 ml
buzlu çay içeren 6 bardak içecek verilmiş, istedikleri kadar içebilecekleri söylenmiştir.
BULGULAR: Kontrol grubu, ortalama 2.0 bardak (50 ml) içecek tüketmiştir. 2. grup, ortalama 2.78 bardak
(69,5 ml) içecek tüketmiştir. 3. grup ise ortalama 4.11 bardak (102,75 ml) içecek tüketmiştir. Kontrol grubu ile
2. grup ve kontrol grubu ile 3. grup arasında yapılan t-testlerinde belirgin fark çıkmıştır (p<0.001).
SONUÇ: Dürtüsel bir süreç olan içecek tüketimi, bilinçaltı düzeyde emosyonel bir uyaran işlemlendiğinde
değişmektedir. Kısa süreli bilinçaltı uyaran esnasında denekler önceden 16 ms boyunca gösterilen emosyonel
yüzleri üst bilinçte fark edememiş olsa da gülen yüz gören grup, kontrol grubuna göre daha çok içecek
tüketmiştir. Uzun süreli bilinçaltı işlemlemede ise, 400 ms boyunca gösterilen gülen yüzler üst bilinçte
tanınmasına karşın, davranışsal tepki cinsiyet ayrımına odaklandığından kognitif süreçte gülen yüzler yine de
bilinçaltı etki yaratmıştır. Bu grupta da gülen yüzler daha fazla içecek tüketimine neden olmuştur.

Modification of incentive motivation in human via subconscious cognitive stimuli
with affective content
OBJECTIVES: Investigating whether beverage consumption behavior can be manipulated through cognitive
processing of emotional stimuli (smiling faces) at the subconscious level.
MATERIALS & METHODS: 27 healthy subjects participated under 3 groups of 9 people. The distribution of
visual stimuli with respect to groups is as follows: 1. Control group: Neutral face for 16 ms, neutral face for 400
ms 2. Subliminal subconscious group: Smiling face for 16 ms, neutral face for 400 ms 3. Supraliminal
subconscious group: Neutral face for 16 ms, smiling face for 400 ms. The pictures are shown within 15 trials,
the subjects are only asked to identify the gender of the person. Prior to the experiment, subjects ate 6 spicy
crackers, and a questionnaire indicated that they were all thirsty. After the experiment, the subjects are offered
6 glasses containing 25 ml of ice tea each.
RESULTS: On the average, the control group consumed 2.0 glasses (50 ml) of the beverage. Group 2
consumed 2.78 glasses (69,25 ml), whereas group 3 consumed 4.11 glasses (102,75 ml). Control group versus
group 2 and control group versus group 3 showed statistically significant difference in t-tests (p<0.001).
CONCLUSION: Beverage consumption driven by implicit motivation can be manipulated through subconscious
cognitive processing of emotional stimuli. When exposed to subliminal visual stimuli which lasted 16 ms, the
subjects could not identify the emotional faces; however, they consumed more beverages if they saw smiling
faces instead of neutral ones. In the processing of supraliminal stimuli which lasted for 400 ms, subconscious
emotional processing occurred because the responses involved gender discrimination rather than emotion
identification. Similarly in this group, beverage consumption is found to be higher than that of the control
group.
