Lipidlar olinishi va xossalariga xos reaksiyalar

Muammoli-modulli o`qitish texnologiyasini qo`llash bilan laboratoriya mashg`ulotlarini o`tkazish (og`zaki bayon qilish, suhbat, munozara, nazariya bo`yicha kollekvium topshirish, sintezni bajarish). Bu texnologiya asosida talabalarda tanqidiy fikrlashni shakllantirish. Olingan birikmalarning fizik-kimyoviy konstantalarin aniqlash.

Mavzu. 1 laboratoriya ishi


1-Tajriba. Glycinning Ninhydrin Bilan Reaktion. Tube 4 Tomchi 1% Li Glycin Erythmasi va 2 Tomchi 0,1% Li Ninhydrin Eritmasidan Tomizing. Aralashmani asta-sekin kukimtir-kizil rank hosil bulguncha kizdiring.

Mavzu. 2 laboratoriya ishi

2-tajriba. Glitsinning formaldegid bilan reaksiyasi.


Probirkaga 5 tomchi 1% li glitsin eritmasidan va 1 tomchi metil kzizil indikatoridan solinadi. Eritma sariq rangga bo‘yaladi (muxit neytral). Olingan aralashmaga teng mikdorda formalin qo‘shiladi va qizil rang xosil bo‘lishiga e’tibor beriladi. Bu reaksiya “formol titrlash” nomi bilan α -aminokislotalarni miqdoriy aniqdash uchun foydalaniladi.
Eritma rangining o‘zgarishi nima bilan bog‘liqligi xaqida xulosa chikdring.

Mavzu. 3 laboratoriya ishi

3-tajriba. Glitsinning nitrit kislota bilan reaksiyasi. Probirkaga 5 tomchi 1% li glitsin eritmasidan va teng xajmda 5% li natriy nitrit eritmasidan solinadi va 2 tomchi konsentrlangan sirka kislota qo‘shib, aralashma oxista chayqatiladi. Gaz ajralib chiqishi kuzatiladi. Bu reaksiya aminokislotalardagi aminoguruxlarni miqdoriy aniqlash uchun ishlatiladi:

Mavzu. 4 laboratoriya ishi


4-tajriba sisteinga rangli reaksiya Probirkaga 5 tomchi 1% li sistein eritmasidan va 2 tomchi 10% li NaOH eritmasidan tomizing. Aralashmani qaynaguncha qizdiring, so‘ng ikki tomchi qo‘rg‘oshin (II) asetatning 10% li eritmasidan tomizing. Probirka tubida qo‘rgoshin sulfidning kul rang qora chukmasi xosil bo‘ladi.


Mavzu. 5 laboratoriya ishi


5-tajriba. Peptid bosta biuret reaksiyasi Probirkaga 5—6 tomchi tuxum oqsili eritmasidan soling va teng xajmda NaOH ning 10% li eritmasidan ko‘shing. Probirka devori buylab 1—2 tomchi CuSO4 eritmasidan tomizing. Qizil pushti rang xosil bo‘ladi. Bu reaksiya peptid bog‘ saqlovchi barcha


birikmalarga sifat reaksiya xisoblanadi.Peptid bog‘ga biuret reaksiyasi.

6-tajriba. Oqsillarning ksantoprotein reaksiyasi.


Probirkaga 10 tomchi tuxum okosili eritmasidan va 2 tomchi konsentrlangan nitrat kislotadan soling. Aralashmani chayqatishni to‘xtatmasdan asta-sekin qizdiring. Eritma va chukma sariq rangga kiradi. Probirkani sovutib, asta-sekin 1—3 tomchi 10% li NaOH eritmasidan tomizing. Tuq zarg‘aldoq rang xosil bo‘ladi. Bu reaksiya aromatik aminokislotalarga (fenilalanin,
tirozin) xos reaksiya bo‘lib, shunday aminokislotalar qoldigani saqdovchi oqsillar xam bu reaksiyani beradi:
