Biochemical analysis of polymorphism and phylogenetic relations as determined by RAPD-PCR of Ammi visnaga and Ammi majus (Umbelliferae)
In this study, it is targeted that determination of antioxidant, antifungal and cytotoxic activities of Ammi majus and Ammi visnaga various extracts and phylogenetic relationship of the species by RAPD-PCR method. Methanol extracts of A. visnaga and A. majus were investigated for their antioxidant capacity by three different assays, namely, the β-carotene linoleic acid systems, DPPH radical scavenging and metal chelating activities. In addition, total phenolics and total flavonoid contents of these methanol extracts were determined. Total phenolics and flavonoid content of A. majus was found to be higher than A. visnaga. However, antioxidant activity of A. visnaga was higher than A. majus in β-carotene linoleic acid systems. The DPPH test system results of A. majus and A. visnaga was found to be similar. Metal chelating potential of A. majus is higher than A. visnaga. Methanol, hexane and chloroform extracts of A. majus and A. visnaga were assayed for antifungal activity against Aspergillus niger and these extracts did not inhibit A. niger. Methanol extract of A. visnaga (root) and A. visnaga (seed) inhibited 50% or over of tumor cells, khellin inhibited other cells 50% or over of tumor cells except DU- 145 and PC-3 cells. Essential oil of A. majus affected only on MCF-7 cell by inhibiting 88% of tumor cells while A. majus (root) methanol extract did not inhibit four tested cells type. But A. visnaga’s phenolic and flavonoid contents are found to be lower in comparison with to those of A. majus. According to this result, phenolic and flovonoid compounds are not responsible for the cytotoxic activity. Plants were comparatively investigated at the molecular level by RAPD technique. Between A. visnaga (Selam) and A. visnaga (Belen) polymorphism value was determined to be 0.041%, between A. visnaga (Selam) and A. majus polymorphism value was determined to be 0.178%, between A. visnaga (Belen) and A. majus polymorphism value was determined to be 0.65%. According to these results, the similarity rate was found as 91% between the samples of A. visnaga (Belen) and A. visnaga (Selam); 89% between the samples of A. majus and A. visnaga (Selam); while it was found as 81% between the samples of A. majus and A. visnaga (Belen). In this study, a dendrogram which shows pylogenetic relationship based on Sorensen index, was prepared regarding to the polymorphic bands, formed by used 20 primers on the samples. These results consisting of 4 primers’ evaluation out of 20 primers, are considered to be repeated with more primers which will form polymorphic bands. Data obtained here are expected to constitute a base for the further studies.
